Abstract:
(EN) In the field of regulation of cell proliferation, a method of inhibiting or preventing cell proliferation is disclosed. Rb, a stable protein encoded by the retinoblastoma gene (RB1), is synthesized at all phases of the cell cycle. Newly synthesized Rb is underphosphorylated during the G0 and G1 phases but is phosphorylated at multiple sites at the G1/S boundary and during S phase. HL-60 cells lose their ability to multiply phosphorylate newly synthesized Rb when induced to terminally differentiate by chemicals such as dimethylsulfoxide, retinoic acid and butyric acid. A method of using these chemicals to inhibit or prevent cell proliferation in cells containing Rb is disclosed.
(FR) Dans le domaine de la régulation de la prolifération de cellules, on décrit un procédé permettant d'inhiber ou empêcher la prolifération des cellules. La Rb, une protéine phosphorylatée codée par le gène du rétinoblastome (RB1) est synthétisée à toutes les phases du cycle de la cellule. La Rb nouvellement synthétisée est sousphosphorylatée durant les phases G0 et G1 mais est phosphorylatée à des sites multiples à la limite G1/S et durant la phase S. Les cellules HL-60 perdent leur capacité d'effectuer une phosphorylation multiple de la Rb nouvellement synthétisée lorsque leur différentiation terminale est induite par des substances chimiques telles que le sulfoxyde diméthylique, l'acide rétinoïque et l'acide butyrique. On décrit un procédé d'utilisation de ces substances chimiques afin d'inhiber ou d'empêcher la prolifération des cellules dans des cellules qui fabriquent le Rb.
