Abstract:
(EN) Proliferating B cells have a cytosolic Transglutaminase (TGase), which exhibits absolute requirement for Ca++ for its activity. The molecular mass of this TGase is 85 kDa. A method for arresting the lymphocytic cell cycle progression in the G1-phase is described by the use of TGase inhibitors, monodansyl cadaverine (MDC) and 1-(5-aminopentyl)-3-phenylthiourea (PPTU). Reversal of this inhibition allows the lymphocytes to enter the S-phase of normal cell cycle progression. A monocytic cell strain exhibits arrest of cell growth and failure to enter S-phase upon treatment with TGase inhibitor.
(FR) Les cellules B prolifératives comportent une transglutaminase (TGase) cytosolique, dont l'activité est tributaire du Ca++. La masse moléculaire de cette TGase est de 85 kDa. Un procédé d'arrêt de la progression du cycle des cellules lymphocytaires dans la phase G1, consiste à utiliser des inhibiteurs de TGase, la cadavérine de monodansyle (CMD) et la 1-(5-aminopentyle)-3-phénylthiourée (PPTU). L'inversion de cette inhibition permet à des lymphocytes de pénétrer dans la phase S de progression du cycle des cellules normales. Une souche de cellules monocytaires permet l'arrêt de la croissance cellulaire et empêche d'entrer dans la phase S grâce au traitement à l'aide d'un inhibiteur de TGase.
