Abstract:
(EN) This invention relates to methods for detecting polynucleotide sequences using nucleic acid hybridization procedures. In particular, the invention concerns the use of oligonucleotides covalently attached to a polymer to increase the rate and extent of capture of specific target nucleic acid contained in a biological sample. In a preferred method, the target nucleic acid in the sample is exposed to a polynucleotide-capturing polymer having multiple oligonucleotide sequences complementary to sequences on the target. After capture of target polynucleotides, the polymer is immobilized (sequestered) onto a solid support through a ligand/receptor combination.
(FR) Cette invention se rapporte à des méthodes permettant de détecter des séquences de polynucléotides à l'aide de procédures d'hybridisation d'acide nucléique. En particulier, l'invention concerne l'utilisation d'oligonucléotides attachés de façon covalente à un polymère afin d'augmenter la cadence et l'ampleur de capture d'un acide nucléique cible spécifique contenu dans un prélèvement biologique. Selon une réalisation préférée, l'acide nucléique dans le prélèvement est exposé à un polymère pouvant capturer un polynucléotide possédant des séquences multiples d'oligonucléotides complémentaires aux séquences sur la cible. Suite à la capture de polynucléotides cibles, le polymère est immobilisé (sequestré) sur un support solide au moyen d'une combinaison ligand/récepteur.
