Abstract:
(EN) Disclosed is a synthetic peptide, useful as an immunocontraceptive agent and for the diagnosis of autoimmune infertility, which corresponds to an autoantigenic epitope of rabbit Sperm Membrane Autoantigen (RSA). The synthetic peptide is selected from the group consisting of the decapeptide NH2-pro-gly-gly-gly-thr-leu-pro-pro-ser-gly-COOH and the antigenic equivalents thereof. Also disclosed is a method of diagnosing autoimmune infertility in a human subject. The method comprises the step of, first, contacting an immune sera sample from the subject with an antigen, the antigen selected from the group consisting of RSA, other proteins which bind selection antibodies which bind to RSA, and derivatives thereof which bind selection antibodies which bind to RSA. The next step is to detect the presence of antibodies from the immune sera sample bound to the antigen, the presence of antibodies bound to the antigen suggesting the subject is afflicted with autoimmune infertility. A preferred group of antigens is the decapeptide NH2-pro-gly-gly-gly-thr-leu-pro-pro-ser-gly-COOH and the antigenic equivalents thereof.
(FR) L'invention concerne un peptide synthétique utile en tant qu'agent immunocontraceptif et pour le diagnostic de l'infécondité auto-immune qui correspond à un épitope autoantigénique de l'autoantigène de la membrane de sperme du lapin (RSA). Le peptide synthétique est sélectionné dans le groupe comprenant le décapeptide NH2-pro-gly-gly-gly-thr-leu-pro-pro-ser-gly-COOH et ses équivalents antigéniques. L'invention concerne également un procédé permettant de diagnostiquer l'infécondité auto-immune chez un être humain. Le procédé consiste en premier lieu à mettre en contact un échantillon de sérum humain du sujet avec un antigène, l'antigène sélectionné dans le groupe comprenant RSA, d'autres protéines qui se lient à des anticorps de sélection qui se lient à RSA, et leurs dérivés qui se lient à des anticorps de sélection qui se lient à RSA. L'étape suivante consiste à détecter la présence d'anticorps dans l'échantillon de sérum humain lié à l'antigène, la présence d'anticorps liés à l'antigène suggérant que le sujet est atteint d'infécondité auto-immune. Un groupe préféré d'antigène et le décapeptide LH2-pro-gly-gly-gly-thr-leu-pro-pro-ser-gly-COOH et ses équivalents antigèniques.
