Abstract:
(EN) A transgene for producing recombinant polypeptides in the milk of transgenic bovine species comprises at least one expression regulation sequence, a secretory DNA sequence encoding a secretory signal which is functional in mammary secretory cells of the bovine species and a recombinant DNA sequence encoding the recombinant polypeptides. A method for producing transgenic bovine species comprises introducing the above transgene into an embryonal target cell of a bovine species, transplanting the transgenic embryonic target cell formed thereby into a recipient bovine parent and identifying at least one female offspring which is capable of producing the recombinant polypeptide in its milk. The invention also includes transgenic bovine species capable of producing recombinant polypeptides in transgenic milk as well as the milk from such transgenic bovine species and food formulations containing one or more recombinant polypeptide. A method for producing transgenic non-human mammals having a desirable phenotype comprises first methylating a transgene followed by introduction into fertilized oocytes. The oocytes are then cultured to form preimplantation embryos. Thereafter, at least one cell is removed from each of the pre-implantation embryos and the DNA digested with a restriction endonuclease capable of cleaving the methylated transgene but incapable of cleaving the unmethylated form of the transgene.
(FR) Un transgène produisant des polypeptides recombinés dans le lait d'espèces bovines transgéniques, comprend au moins une séquence de régulation d'expression, une séquence d'ADN sécrétoire codant un signal sécrétoire fonctionnel dans des cellules sécrétoires mammifères d'espèces bovines, ainsi qu'une séquence d'ADN recombiné codant les polypeptides recombinés. Un procédé de production d'espèces bovines transgéniques consiste à introduire le transgène précité dans une cellule cible embryonnaire d'une espèce bovine, à transplanter la cellule cible embryonnaire transgénique, ainsi transformée en parent bovin récepteur, et à identifier au moins un déscendant femelle capable de produire le polypeptide recombiné dans son lait. L'invention comprend également des espèces bovines transgéniques capables de produire des polypeptides recombinés dans du lait transgénique ainsi que le lait provenant desdites espèces bovines transgéniques, et des formulations alimentaires contenant un ou plusieurs polypeptides recombinés. Un procédé de production de mammifères non humains transgéniques présentant le phénotype recherché, consiste premièrement à méthyler un transgène puis à l'introduire dans des ovocytes fécondés. Ensuite, on met en culture les ovocytes afin de former des embryons de pré-implantation. Ensuite, au moins une cellule est retirée de chacun des embryons de pré-implantation, et l'ADN est digéré à l'aide d'une endonucléase de restriction capable de cliver le transgène méthylé mais incapable de cliver la forme non méthylée du transgène.
