Abstract:
(EN) A DNA fragment distinct from the epidermal growth factor receptor (EGF-R) and $i(erb)B-2 genes was detected by reduced stringency hybridization of v-$i(erb)B to normal genomic human DNA. Characterization of the cloned DNA fragment mapped the region of v-$i(erb)B homology to three exons with closest homology of 64 % and 67 % to a contiguous region within the tyrosine kinase domains of theEGF-R and $i(erb)B-2 proteins, respectively. cDNA cloning revealed a predicted 148 kd transmembrane polypeptide with structural features identifying it as a member of the $i(erb)B family, prompting designation of the new gene as $i(erb)B-3. It was mapped to human chromosome 12q11-13 and was shown to be expressed as 6.2 kb transcript in a variety of normal tissues of epithelial origin. Markedly elevated $i(erb)B-3 mRNA levels were demonstrated in certain human mammary tumor cell lines. These findings indicate that increased $i(erb)B-3 expression, as in the case of EGF-R and $i(erb)B-2, plays a role in some human malignancies.
(FR) Un fragment ADN distinct des gènes $i(erb)B-2 et du récepteur du facteur de croissance épidermique (EGF-R) a été détecté par hybridation de force réduite de v-$i(erb)B sur l'ADN humain génomique normal. La caractérisation du fragment ADN cloné a topographié la région d'homologie de v-$i(erb)B sur trois exons avec une homologie la plus proche de 64 % et 67 % sur une région contigue à l'intérieur des domaines de la kinase de tyrosine des protéines EGF-R et $i(erb)B-2, respectivement. Le clonage de l'ADN complémentaire a révélé un polypeptide transmembranaire 148 kd de prédiction avec des caractéristiques de structure l'identifiant comme étant un élément de la famille $i(erb)-B, suggérant de désigner le nouveau gène $i(erb)B-3. Il a été topographié sur le chromosome humain 12q11-13 et a été exprimé comme transcription 6,2 kb dans une variété de tissus normaux d'origine épithéliale. Des niveaux d'ARN messager dans $i(erb)B-3 particulièrement élevés ont été trouvés dans certaines lignées cellulaires de tumeurs mammaires humaines. Ces découvertes indiquent que l'expression de $i(erb)B-3 augmenté, comme dans le cas de EGF-R et $i(erb)B-2, joue un rôle dans certaines malignités humaines.
