Abstract:
(EN) A method of isolating fetal nucleated cells, particularly fetal nucleated erythrocytes, from a maternal blood sample, by means of an antigen present on the cell surface of the fetal erythrocytes. A method of detecting fetal DNA of interest, which is a gene or gene portion associated with a disease or condition, a chromosomal abnormality or sex-specific DNA, in a maternal blood sample. The presence or absence, as well as the quantity of fetal DNA of interest in a maternal sample can be determined. The claimed method of detection can be used prenatally or postnatally and is particularly useful because it is noninvasive and can be carried out early in pregnancy.
(FR) Procédé d'isolation de cellules f÷tales nucléées, notamment d'erythrocytes f÷taux nucléés, à partir d'un échantillon de sang maternel, au moyen d'un antigène présent sur la surface cellulaire des erythrocytes f÷taux, et procédé de détection d'ADN f÷tal d'intérêt, lequel est un gène ou une partie de gène associée à une maladie ou à un état, à une anomalie chromosomique ou à de l'ADN spécifique au sexe, dans un échantillon de sang maternel. La présence ou l'absence, ainsi que la quantité d'ADN f÷tale d'intérêt dans un échantillon maternel, peuvent être déterminées. On peut utiliser le procédé de détection de l'invention en période pré-natale ou post-natale, et il est particulièrement utile du fait qu'il n'est pas envahissant et qu'il peut être mis en ÷uvre précocement dans la grossesse.
