Abstract:
(EN) A process for the production of a heterologous polypeptide or protein molecule through expression in a eucaryotic host cell, comprising the steps: (a) transforming said host cell by introducing therein an expression vector, said vector comprising an upstream regulatory sequence, a first DNA fragment encoding said molecule and positioned downstream of said regulatory sequence, which is capable of initiating RNA transcription of said DNA fragment, and further comprising a second DNA fragment with signals for efficient RNA processing located downstream of said first DNA fragment; (b) culturing the transformed host cell in a suitable medium under conditions allowing expression of the desired molecule; and (c) recovering the molecule resulting from step (b) from the culture; host cells transformed by introducing therein such an expression vector; and polypeptide and protein molecules prepared by such process.
(FR) Procédé de production d'une molécule polypeptidique ou protéique hétérologue par expression dans une cellule hôte eucaryote, comprenant les étapes consistant (a) à transformer ladite cellule hôte par introduction dans celle-ci d'un vecteur d'expression, ledit vecteur comprenant une séquence régulatrice amont, d'un premier fragment d'ADN codant ladite molécule et positionné en aval de ladite séquence régulatrice, lequel est capable d'amorcer une transcription d'ARN dudit fragment d'ADN, et comprenant également un second fragment d'ADN présentant des signaux de traitement efficace d'ARN, le deuxième fragment se trouvant en aval dudit premier fragment d'ADN; (b) à cultiver la cellule hôte transformée dans un milieu approprié dans des conditions permettant l'expression de la molécule voulue; et (c) à récupérer la molécule résultant de l'étape (b) du bouillon de culture; les cellules hôtes transformées par introduction dans celles-ci dudit vecteur d'expression; et les molécules polypeptidiques et protéiques préparées selon le procédé.
