Abstract:
(EN) Recombinant protein complexes having human Factor VIII:C activity are expressed in a eukaryotic host cell by transforming the host cell with first and second expression cassettes encoding a first polypeptide substantially homologous to human Factor VIII:C A domain and a second polypeptide substantially homologous to human Factor VIII:C C domain, respectively. In the present invention, the first polypeptide may be extended having at its C-terminal a human Factor VIII:C B domain N-terminal peptide, a polypeptide spacer of 3-40 amino acids, and a human Factor VIII:C B domain C-terminal peptide. Expression of the second polypeptide is improved by employing an $g(a)1-antitrypsin signal sequence.
(FR) On a exprimé des complexes de protéines recombinées présentant l'activité du facteur VIII:C humain, dans une cellule hôte eucaryote, par transformation de la cellule hôte à l'aide de première et seconde cassettes d'expression codant un premier polypeptide sensiblement homologue au domaine A du facteur VIII:C humain, et un second polypeptide sensiblement homologue au domaine C du facteur VIII:C humain, respectivement. Selon l'invention, on peut prolonger le premier polypeptide afin qu'il présente au niveau de sa terminaison C un peptide à terminaison N du domaine B du facteur VIII:C humain, un élément d'espacement polypeptidique de 3 à 40 acides aminés, ainsi qu'un peptide à terminaison C du domaine B du facteur VIII:C humain. On a amélioré l'expression du second polypeptide en utilisant une séquence de signaux de $g(a)1-antitrypsine.
