Abstract:
(EN) A mutation defining a gene for the N$g(a)-acetyltransferase of yeast has been identified and cloned. Mutations in this gene have been constructed and used to produce cells which are substantially incapable of catalyzing N$g(a)-acetylation of protein. Such cells are valuable tools for determining the amino acid sequence of uncharacterized proteins. Such cells are also valuable tools for expressing a recombinant protein lacking an acetyl group at its $g(a)-amino group.
(FR) On a identifié et cloné une mutation définissant un gène destiné à la N$g(a)-acétyltransférase de levure. Les mutations se produisant dans ce gène ont été élaborées et utilisées afin de produire des cellules incapables de catalyser la N$g(a)-acétylation de protéine. Lesdites cellules constituent des outils de valeur permettant de déterminer la séquence d'acides aminés de protéines non caractérisées. Lesdites cellules constituent également des outils de valeur permettant d'exprimer une protéine recombinée dépourvue d'un groupe acétyle au niveau de son groupe $g(a)-amino.
