Abstract:
(EN) There is described a method, for obtaining a eukaryotic cell containing in its DNA multiple copies of a GS gene, comprising: transforming a eukaryotic glutamine auxotroph with a GS gene; selecting transformant cells containing the GS gene; and culturing the selected transformant cells in a medium which lacks glutamine or in which the amount of glutamine is progressively depleted, the GS gene being of a character such that, or the conditions employed during the culturing step being such that, the GS gene is so weakly transcribed that cells in which the GS gene has been amplified are selected.
(FR) Description d'une méthode d'obtention d'une cellule eukariotique contenant dans son ADN des copies multiples d'un gène de synthétase de glutamine (GS), comprenant : la transformation d'un auxotrophe glutaminique eukariotique par un gène de synthétase glutaminique GS; la sélection des cellules transformantes contenant le gène de synthétase glutaminique GS; la culture des cellules transformantes selectionnées dans un milieu manquant de glutamine ou dans lequel la quantité de glutamine est progressivement épuisée, les caractéristiques du gène de synthétase glutaminique et les conditions appliquées pendant le stade de culture étant telles que le gène de synthétase glutaminique GS est transcrit si faiblement que l'on sélectionne des cellules dans lesquelles il a été amplifié.
