Abstract:
(EN) The invention is based on the finding that furin belongs to a family of endoproteolytically active enzymes and relates to a process for the $i(in vitro) cleavage of a protein by treating the protein in the presence of Ca2+ ions with furin or a furin-like enzyme, or an endoproteolytically active fragment, derivative or fusion protein of furin or furin-like enzyme. The invention can be used for the (micro)biological production of a protein by culturing genetically engineered cells expressing a proform of the protein as well as furin or a furin-like enzyme and isolating the protein formed. The invention also relates to a pharmaceutical composition comprising one or more pharmaceutically acceptable carriers, diluents or adjuvants, as well as an endoproteolytically active amount of furin or a furin-like enzyme, or a fragment or derivative of furin or furin-like enzyme having an endoproteolytic activity.
(FR) L'invention se fonde sur la découverte que la furine fait partie d'une famille d'enzymes à activité endoprotéolytique, et concerne un procédé de segmentation $i(in vitro) d'une protéine par le traitement de la protéine, en présence d'ions de Ca2+, avec de la furine ou une enzyme semblable à la furine, ou avec un fragment, un dérivé ou une protéine de fusion à activité endoprotéolytique de furine ou d'une enzyme semblable à la furine. L'invention peut être utilisée lors de la production (micro)biologique d'une protéine au moyen de la culture de cellules modifiées par des techniques de génie génétique qui expriment un substitut de la protéine ainsi que la furine ou l'enzyme semblable à la furine, et qui isolent la protéine ainsi formée. L'invention concerne également une préparation pharmaceutique comportant un ou plusieurs porteurs, diluants ou adjuvants acceptables en pharmacologie, ainsi qu'une quantité à activité endoprotéolytique de furine ou d'une enzyme semblable à la furine, ou un fragment ou un dérivé de furine ou d'une enzyme semblable à la furine présentant une activité endoprotéolytique.
