Abstract:
(EN) Disclosed herein is a method for cryopreserving musculoskeletal tissues, such as ligaments, tendons and cartilage, by placing such tissue in contact with a composition containing a cryopreserving agent comprising a cell-penetrating organic solute, which is preferably dimethylsulfoxide, and a glycosaminoglycan, which is preferably chondroitin sulphate, in an amount sufficient to cryopreserve the musculoskeletal tissue. The addition of a glycosaminoglycan to a cryopreserving agent comprising a cell-penetrating organic solute permits a broad range of cooling rates to be employed, rather than the very narrow ranges which are employed using a cryopreserving agent comprising a cell-penetrating organic solvent without the glycosaminoglycan. Also disclosed are a freezing schedule designed to maximize retention of tissue cell viability and biomechanical properties during and after the freezing process, and a thawing schedule which maximizes cell viability.
(FR) On décrit un procédé de cryoconservation des tissus musculo-squelettiques, tels que les ligaments, les tendons et le cartilage, consistant à mettre ces tissus en contact avec une préparation contenant un agent de cryoconservation comprenant un soluté organique pouvant pénétrer les cellules, qui est de préférence le diméthylsulfoxyde, et un glycosaminoglycan, qui est de préférence le dermatane-sulfate, en quantité suffisante pour cryoconserver le tissu musculo-squelettique. L'ajout d'un glycosaminoglycan à un agent de cryoconservation comprenant un soluté organique pouvant pénétrer les cellules permet l'utilisation d'une pluralité de vitesses de refroidissement, au lieu du nombre très limité de celles-ci que l'on utilise avec un agent de cryoconservation qui comprend un soluté organique pouvant pénétrer les cellules, mais qui ne contient pas le glycosaminoglycan. On décrit également un programme de congélation conçu pour maximiser la rétention de la viabilité des cellules tissulaires et des caractéristiques biomécaniques pendant et après le processus de congélation, ainsi qu'un programme de dégel maximisant la viabilité cellulaire.
