A method for controlling and modifying biopolymer synthesis by manipulation of the genetics and enzymology of synthesis of polyhydroxybutyrate (PHB) and polyhydroxyalkanoate (PHA) polyesters at the molecular level in procaryotic and eukaryotic cells, especially plants. Examples demonstrate the isolation, characterization, and expression of the genes involved in the production of PHB and PHA polymers. Genes encoding the enzymes in the PHB and PHA synthetic pathway (beta-ketothiolase, acetoacetyl-CoA reductase and PHB polymerase or PHA polymerase) from $i(Zoogloea ramigera) strain I-16-M, $i(Alcaligenes eutrophus), $i(Nocardia salmonicolur), and $i(Pseudomonas oleovarans) were identified or isolated and expressed in a non-PHB producing organism, $i(E. coli). Specific modifications to the polymers include variation in the chain length of the polymers and incorporation of different monomers into the polymers to produce co-polymers with different physical properties.

Procédé de régulation et de modification de la synthèse de biopolymères par manipulation génétique et enzymologie de la synthèse de polyesters de polyhydroxybutyrate (PHB) et de polyhydroxyalkanoate (PHA) au niveau moléculaire dans des cellules procaryotiques et eukaryotiques, spécialement chez les plantes. Des exemples montrent l'isolement, la caractérisation et l'expression des gènes impliqués dans la production des polymères PHB et PHA. Des gènes codant les enzymes dans le chemin synthétique de PHB et de PHA (bêta-cétothiolase, réductase acétoacétyle-CoA et polymérase PHB ou polymérase PHA) à partir de la souche $i(Zoogloea ramigera) I-16M $i(Alcaligenes eutrophus), $i(Nocardia salmonicolur), et $i(Pseudomonas oleovarans) ont été identifiés ou isolés et exprimés dans un organisme ne produisant pas de PHB, $i(E. coli). Des modifications spécifiques apportées aux polymères comprennent la variation dans la longueur de la chaîne des polymères ainsi que l'incorporation de différents monomères dans les polymères pour produire des copolymères ayant des propriétés physiques différentes.
