Abstract

The present invention relates to novel antibodies reactive with human carcinoma cells. More particularly, the antibodies of the invention include: a murine monoclonal antibody, BR96; a human/murine chimeric antibody, ChiBR96; and a F(ab')2 fragment of BR96. These antibodies are reactive with a cell membrane antigen on the surface of human carcinomas. The antibodies display a high degree of selectivity for carcinoma cells and possess the ability to mediate ADCC and CDC activity. In addition, the antibodies of the invention internalize within the carcinoma cells to which they bind and are therefore particularly useful for therapeutic applications, for example, as the antibody component of antibody-drug or antibody-toxin conjugates. The antibodies also have a unique feature in that they are cytotoxic when used in the unmodified form, at specified concentrations.

L'invention concerne de nouveaux anticorps réagissant avec des cellules de carcinome humain. Plus particulièrement, les anticorps de l'invention comprennent: un anticorps monoclonal murin, le BR96; un anticorps chimérique humain/murin, le ChiBR96; et un fragment F(ab')2 de BR96. Ces anticorps réagissent avec un antigène de membrane cellulaire présent sur la surface des carcinomes humains. Les anticorps présentent un degré élevé de sélectivité pour les cellules de carcinome et possèdent la capacité de médiation de l'activité ADCC et CDC. En outre, les anticorps de l'invention s'internalisent à l'intérieur des cellules de carcinome auxquelles ils se lient et sont donc particulièrement utiles aux applications thérapeutiques, par exemple, en tant que composant anticorps des conjugués anticorps-médicament et anticorps-toxine. Les anticorps possèdent également une caractéristique unique en ce qu'ils sont cytotoxiques lorsqu'ils sont utilisés en forme inchangée, à des concentrations spécifiées.
