Abstract

This invention provides a method of assessing glycosylated haemoglobin in a sample, said method comprising the steps of: (a) optionally haemolysing the sample to liberate any cell bound haemoglobin; (b) contacting the said sample or haemoglobin recovered from said sample according to step (c) below with signal-forming molecules comprising a conjugate of one or more dihydroxyboryl residues or salts thereof, linked to a signal-forming label; (c) separating glycosylated and non-glycosylated haemoglobin and any molecules bound thereto, from the sample or from the reaction mixture of step (b) above; and (d) assessing the said signal forming molecules which have bound to the separated haemoglobin, and/or any non-haemoglobin bound signal-forming molecules; and an analytical test kit for use in accordance with the method of the invention. The new assay of the invention is particularly useful for the $i(in vitro) diagnosis and monitoring of diabetes mellitus.

Le procédé de mesure de l'hémoglobine glucosylée présente dans un échantillon consiste: (a) à effectuer en option l'hémolyse de l'échantillon pour libérer l'hémoglobine liée aux globules; (b) à mettre en contact cet échantillon ou l'hémoglobine extraite de l'échantillon selon l'étape (c) ci-dessous avec des molécules formant signal comprenant un conjugué d'un ou de plusieurs résidus de dihydroxyboryle ou de sels de ces résidus, reliés à une étiquette formant signal; (c) à séparer l'hémoglobine glucosylée et non glucosylée et toutes les molécules liées à elle, de l'échantillon ou du mélange de réaction de l'étage (b) ci-dessus; et (d) à mesurer la quantité de ces molécules formant signal, qui se sont liées à l'hémoglobine séparée, et/ou toutes les molécules formant signal non liées à l'hémoglobine. L'invention se rapporte également à un kit de test analytique à utiliser conjointement au procédé décrit ci-dessus. Ce nouveau procédé d'analyse est particulièrement utile pour le diagnostic et le contrôle $i(in vitro) du diabète sucré.
