Abstract

A process for preparing microspheres using very cold temperatures to freeze polymer-biologically active agent mixtures into polymeric microspheres with very high retention of biological activity and material. Polymer is dissolved in a solvent together with an active agent that can be either dissolved in the solvent or dispersed in the solvent in the form of microparticles. The polymer/active agent mixture is atomized into a vessel containing a liquid non-solvent, alone or frozen and overlayered with a liquified gas, at a temperature below the freezing point of the polymer/active agent solution. The cold liquified gas or liquid immediately freezes the polymer droplets. As the droplets and non-solvent for the polymer is warmed, the solvent in the droplets thaws and is extracted into the non-solvent, resulting in hardened microspheres.

L'invention concerne un procédé de préparation de microsphères à des températures très basses pour congeler des mélanges de polymère et d'agent biologiquement actif dans des microsphères polymères avec une très grande rétention de l'activité biologique, ainsi que la matière utilisée dans ce procédé. Un polymère est dissous dans un solvant avec un agent actif qui peut être soit dissous dans le solvant soit dispersé dans le solvant sous la forme de microparticules. Le mélange polymère/agent actif est atomisé dans un récipient contenant un liquide non solvant seul ou congelé et recouvert par une couche de gaz liquéfié à une température inférieure au point de congélation de la solution polymère/agent actif. Le liquide ou gaz liquéfié froid congèle immédiatement les gouttelettes de polymère. Au fur et à mesure que les gouttelettes et le non-solvant pour le polymère sont chauffés, le solvant dans les gouttelettes se décongèle et est extrait des gouttelettes pour entrer dans le non-solvant liquide et former des microsphères durcies.
