Abstract
(EN)
An improved method for testing for the presence of a particular microorganism and/or group of microorganisms characterized by a particular enzyme is provided. The method uses a dye-forming and/or fluorogenic-generating substrate throughout a polymer matrix or on the surface of a solid support member that forms a colored precipitate or fluorescent by-product when cleaved by the enzyme. The precipitate is concentrated on the polymer matrix and/or solid support member to create a visible reaction product, whereas the fluorescent by-product will normally be dispersed into the surrounding medium to some extent. The amount of dye-forming substrate needed is independent of the sample size. The present invention further comprises an enzyme indicator device comprising a dye-forming and/or fluorogenic-generating substrate throughout a polymer system or on the surface of a solid support member.

Un procédé amélioré permet de détecter la présence d'un microorganisme particulier et/ou d'un groupe de microorganismes caractérisé par un enzyme particulier. Le procédé utilise un substrat de formation de colorant et/ou générateur de fluorogène dans une matrice polymère ou à la surface d'un organe de support solide qui forme un précipité coloré ou un sous-produit fluorescent lorsqu'il est clivé par l'enzyme. Le précipité est concentré sur la matrice polymère et/ou sur l'organe de support solide pour créer un produit de réaction visible, tandis que le sous-produit fluorescent se dispersera normalement dans le milieu environnant, dans une certaine mesure. La quantité de substrat formant colorant nécessaire dépend de la taille de l'échantillon. La présente invention concerne en outre un dispositif indicateur enzymatique comprenant un substrat de formation de colorant et/ou de génération de fluorogène dans tout un système polymère ou sur la surface d'un organe de support solide.
