A method and apparatus (320, 340, 360) for automatically and rapidly retrieving, counting and/or analyzing at least one selected population of cells or formed bodies, such as a white blood cell population and at least one subset thereof of a whole blood sample (322, 342) or portion thereof. A volume of a biological medium containing the white blood cells is prepared and at least one reactant (326, 346) specific or preferential at least to some selected biological cells is introduced thereto and rapidly mixed (384) for a short period of time. The opacity and/or volume parameter of the cells can be modified and the mixture is then counted and analyzed (332, 352, 416, 422) in one or more steps to obtain the desired white blood cell population and subset analysis. The biological sample can be a whole blood sample and the reactant can include or be a lyse or a monoclonal antibody bound to microspheres, which will bind to specific ones of the cells or a combination of lyse and microspheres with antibody bound thereto. The microspheres can be magnetic and the bound cells can be magnetically removed for retrieving and analyzing the remaining blood cell population.

L'invention concerne un procédé et un appareil (320, 340, 360) permettant la récupération, le comptage et/ou l'analyse automatique et rapide d'au moins une population sélectionnée de cellules ou de corps formés, tels qu'une population de globules blancs ainsi que d'au moins un sous ensemble de cette dernière d'un échantillon de sang entier (322, 342) ou d'une partie de celui-ci. On prépare un volume d'un milieu biologique contenant les globules blancs, on y introduit au moins un réactif (326, 346) spécifique ou préférentiel au moins à plusieurs cellules biologiques sélectionnées, puis on les mélange (384) pendant une courte période de temps. On peut modifier le paramètre d'opacité et/ou de volume des cellules, puis on compte et on analyse (332, 352, 416, 422) le mélange en une ou plusieurs étapes afin d'obtenir l'analyse de la population et du sous-ensemble de globules blancs. L'échantillon biologique peut être un échantillon de sang entier et le réactif peut inclure ou être un lysat ou un anticorps monoclonal lié à des microsphères, se liant à des cellules spécifiques ou à une combinaison de lysat et de microsphères auxquelles des anticorps sont liés. Les microsphères peuvent être magnétiques et les cellules liées peuvent être magnétiquement retirées afin de récupérer et d'analyser la population de cellules sanguines restante.
