Abstract:
(EN) Method for diagnosing infections by agents with distinct lipopolysaccharide antigens in which clinical samples are applied to a support which absorbs the lipopolysaccharide antigen and then the support, carrying the antigen, is treated with a solution of antibody (especially monoclonal antibody) which has affinity for the antigen and a means for rendering it detectable, especially a radio-label. The absorbing support's preferred material is nitrocellulose, and its preferred form is a membrane. Accuracy and efficiency are improved by treating samples, before application to the absorbing support, with a proteolytic enzyme which is later deactivated. Treatment of the support or membrane with a solution of high-protein content, after it has absorbed the antigen, gives further improvement. The method is easily used by the immune blot technique and can be used as a genus-specific method for diagnosing Chlamydia (C.trachomatis and C.psittaci).
(FR) Procédé permettant de diagnostiquer des infections par des agents ayant des antigènes de lipopolysaccharide différents, dans lequel on applique des échantillons cliniques à un support absorbant l'antigène de lipopolysaccharide, le support portant l'antigène étant ensuite traité avec une solution d'anticorps (notamment d'anticorps monoclonaux) ayant une affinité à l'égard de l'antigène et un moyen permettant de le rendre détectable, notamment une marque radioactive. La matière préférée du support absorbant est la nitrocellulose et sa forme préférée est une membrane. On augmente la précision et l'efficacité par traitement des échantillons, avant application au support absorbant, avec une enzyme protéolytique qui est ensuite désactivée. Le traitement du support ou de la membrane avec une solution à teneur élevée en protéine, après l'absorption de l'antigène par le support ou la membrane, donne un résultat encore meilleur. Le titrage par immunotransfert utilise aisément le procédé qui peut être employé comme procédé spécifique à un gène permettant de diagnostiquer le chlamydia (c.trachomatis et C.psittaci).
