Abstract:
(EN) Protein analogues of tissue plasminogen activator (tPA). The analogues exhibit the proteolytic function of natural tPA and optionally fibrin binding activity, but are molecules, generally of lower molecular weight than natural tPA, designed for efficient expression in prokaryotic host cell systems. The analogues can comprises a catalytic fragment of tPA or a catalytic fragment of tPA linked to a polypeptide which stabilizes the catalytic fragment, provides for efficient expression of the fragment or confers a fibrin binding capability. Fibrin binding polypeptides can be polypeptide fragments derived from tPA which embody the fibrin binding domain(s) of natural tPA or they can be an exogenous (non-tPA) polypeptides of eukaryotic or prokaryotic origin which exhibit fibrin binding affinity such as the antigen binding fragment of an antifibrin immunoglobulin or the B domain of protein A. Genetic constructs for expression of the analogues are also provided.
(FR) Les analogues de protéine d'un activateur de plasminogène tissulaire (dPA) décrits présentent la fonction protéolytique du tPA naturel et éventuellement une activité de liaison de fibrine, mais sont des molécules en général d'un poids moléculaire plus faible que celui du tPA naturel, et sont conçues pour l'expression efficiente dans des systèmes de cellules hôtes prokaryotiques. Les analogues peuvent comprendre un fragment catalytique de tPA ou un fragment catalytique de tPA lié à un polypeptide qui stabilise le fragment catalytique, assure l'expression efficiente du fragment ou lui donne la capacité de laison de fibrine. Les polypeptides de liaison de fibrine peuvent être des fragments de polypeptide dérivés de tPA qui constituent le ou les domaines de liaison de fibrine du tPA naturel ou peuvent être des polypeptides hexogènes (non-tPA) d'origine eukaryote ou prokaryote qui présentent une affinité de laison de fibrine telle que le fragment de liaison d'un antigène d'une immunoglobuline antifibrine ou le domaine B de la protéine A. Des constructions génétiques pour l'expression des analogues sont également décrites.
