(EN)Method of determining a substrate in a sample which comprises converting the substrate to a product in a first stage of a cyclic reaction sequence and converting the product back to the substrate in a second reaction stage of the cyclic reaction sequence. At least one of the first and second reaction stages is enzyme catalyzed. Simultaneously with either the first reaction stage or the second reaction stage an indicating substrate is convened to an indicating product in a non-reversible indicator reaction. After this the loss of indicating substract or gain of indicating product may be determined. The method is particularly efficacious in enzyme immunoassay determinations which include homogeneous enzyme immunoassay and enzyme linked immunoassay wherein the free or bound enzyme-ligand or enzyme antibody conjugate is to be assayed by the method of the invention.
(FR)Procede de determination d"un substrat dans un echantillon comprenant la conversion du substrat en un produit dans une premiere etape d"une sequence cyclique de reaction et la reconversion du produit en substrat dans une deuxieme etape de reaction de la sequence cyclique de reaction. Au moins une etape reactionnelle, soit la premiere soit la deuxieme, est catalysee par une enzyme. Simultanement, soit a la premiere etape de reaction soit a la deuxieme, un substrat d"indication est converti en un produit d"indication dans une reaction d"indicateur non reversible. Ensuite la perte d"un substrat d"indication ou le gain d"un produit d"indication peut etre determinee. Le procede est particulierement efficace lors des determinations d"enzymes par analyse immunologique incluant des analyses immunologiques d"enzyme homogenes et des analyses immunologiques enzymatiques dans lesquelles le liant d"enzyme libre ou lie ou le conjugue d"anticorps enzymatique doit etre analyse par le procede selon l"invention.
