(FR) METHODE DE SEPARATION DES CELLULES
Abstract:
(EN) A method of cell separation by agglutination using lectins or agglutinins which selectively bind to cells devoid of graft-versus-host-activity and suitable for transplantation without undesirable side effects. The method comprises preparing a mixed cell suspension in a suitable medium, contacting this mixed cell suspension with a lectin, which may be peanut agglutinin or soybean agglutinin, or agglutinin that selectively binds to terminal galactose residues on the cell surface and forms aggregates of cells, separating the cells from the unaggregated cells, and contacting the separated cell aggregates with D-galactose solution in order to obtain single cells suitable for injection.
(FR) Une methode de separation de cellules par agglutination utilisant des lectines ou des agglutines qui se lient selectivement aux cellules denuee d"une activite hote contre greffe et appropriee pour une transplantation sans effets secondaires indesirables. La methode consiste a preparer une suspension de cellules melangees dans un milieu approprie, a mettre en contact cette suspension de cellules melangees avec une lectine qui peut etre de l"agglutine d"arachides ou de soja, ou de l"agglutine qui se lie selectivement au residu final de galactose sur la surface des cellules et forme des agregats de cellules, a separer les cellules non agregees, et a mettre en contact les agregats de cellules separees avec une solution de D-galactose de maniere a obtenir des cellules individuelles appropriees a l"injection.
