(EN)An immunochemical method for detecting prostatic acid phosphatases has been developed which is useful in clinical laboratory testing for prostate cancer and in enzyme and protein determinations. An isoenzymatically homogeneous prostatic acid phosphatase antigen is prepared by isolating purified acid phosphatase from cancerous human prostate tissue. The antigen stimulates the production of diagnostic antibodies which are highly specific, do not cross-react with other acid phosphatase isoenzymes and can be used in countercurrent immuno-electrophoresis (showing a sensitivity of 0.4 I.U. of enzyme activity or 20 ng/ml enzyme protein) and in a solid phase fluorescent immunoassay (showing a sensitivity of 60 pg/ml enzyme protein with excitation of the alpha-naphthol hydrolysis product at 340 nm and emission at 465 nm) with no false positives and few false negatives in diagnosing cancer of the prostate.
(FR)La methode immunochimique pour detecter les phosphatases prostatiques acides est utilisee en laboratoire clinique pour la recherche du cancer de la prostate et pour des determinations d"enzymes et de proteines. On prepare un antigene de phosphatase prostatique, acide, isoenzymatique, en isolant une phosphatase acide, purifiee, a partir de tissus humains cancereux de la prostate. L"antigene stimule la production d"anticorps de diagnostic qui sont tres specifiques, n"interferent pas avec d"autres isoenzymes de phosphatase acide et peuvent etre utilises dans des immuno-electrophoreses a contre-courant (montrant une sensibilite de 0,4 I.U. d"activite enzymatique ou 20 ng/ml d"enzyme proteinique) et dans une analyse immunologique de fluorescence en phase solide (montrant une sensibilite de 60 pg/ml d"enzyme proteinique avec excitation du produit d"hydrolyse de l"alpha-naphtol a 340 nm et emission a 465 nm) avec aucun resultat positif faux et peu de resultats negatifs faux dans le diagnostic du cancer de la prostate.
