(FR) PROCEDE DE DETERMINATION DE SUBSTANCES BIOLOGIQUES PAR DIFFUSION DANS UNE MATRICE POREUSE OU PAR ELECTROPHORESE
Abstract:
(EN) A method for the determination of the quantity and type of a biological component A in a sample which is added to a basin in a liquid-saturated, porous matrix covering a thin layer of a biologically active component B bound to the solid surface of a carrier. A further biologically active component C included in the matrix is capable of reacting biospecifically with component A when the sample is allowed to diffuse or migrate electrophoretically in the matrix forming a zone containing a precipitate of components A and C. This precipitate will adsorb with biospecificity to component B, bound to the solid surface of the carrier and is visualized in a suitable manner on the surface after removal of the said porous matrix, eg. as a change in the surface tension angle observed with the aid of vapour condensation on the surface. Components B and C or B and A may be the same as long as the remaining component is capable of reacting with both the others. The components A, B and C may be antigens and antibodies. The carrier is suitably made of plastic material.
(FR) Procede de determination de la quantite et du type d"un composant biologique A dans un echantillon qui est dispose dans une cuvette formee par une matrice poreuse saturee de liquide, recouvrant une fine couche d"un composant biologiquement actif B lie a la surface solide d"un support. Un autre composant biologiquement actif C inclus dans la matrice est capable de reagir biospecifiquement avec le composant A lorsque l"echantillon peut diffuser ou migrer par voie electrophoretique dans la matrice, formant une zone contenant un precipite des composants A et C. Ce precipite s"absorbe de maniere biospecifique sur le composant B, lie a la surface solide du support et est rendu visible de maniere appropriee sur la surface apres enlevement de ladite matrice poreuse, par exemple par un changement de l"angle de tension de surface observe a l"aide de la condensation de vapeur sur la surface. Les composants B et C ou B et A peuvent etre les memes pour autant que le composant restant puisse reagir avec les deux autres. Les composants A, B et C peuvent etre des antigenes et des anticorps. Une matiere plastique peut constituer un materiau approprie par le support.
