Abstract

This disclosure concerns compositions and methods for targeting peptides, polypeptides, and proteins to plastids of plastid-containing cells. In some embodiments, the disclosure concerns a transgenic plant cell comprising a genomic, chromosomal nucleic acid molecule comprising a polynucleotide operably linked to a promoter that is functional in a plant cell, the polynucleotide encoding a chimeric polypeptide consisting of a synthetic chloroplast transit peptide (CTP) that is less than 80 amino acids in length and a polypeptide of interest, the amino acid sequence of the synthetic CTP consisting of: a contiguous amino acid sequence of a first CTP from a first Brassica plant 3-enolpyruvylshikimate-5-phosphate synthase (EPSPS) that is between 25 and 41 amino acids in length; and a second contiguous amino acid sequence of a second, different CTP from a second Brassica plant EPSPS, wherein the second contiguous amino acid sequence of the second, different CTP does not overlap with the contiguous amino acid sequence of the first CTP in the comparison window of an alignment of the first and second Brassica plant EPSPS CTPs, and wherein the plant cell comprises the polypeptide of interest in a chloroplast of the plant cell.
