Abstract:
(ES) Se describen mutantes de la alfa-amilasa novedosos, derivados de las secuencias de ADN de alfa-amilasas que se encuentran en la naturaleza o recombinantes. Las alfa-amilasas mutantes, en general, se obtienen por modificaciones in vitro de una secuencia de ADN precursor que codifica para la alfa-amilasa que se encuentra en la naturaleza o recombinante para generar la sustitución (reemplazo) o supresión de uno o más aminoácidos residuales oxidables en la secuencia de aminoácido de una alfa-amilasa precursora. Tales alfa-amilasas mutantes tiene estabilidad oxidativa alterada y/o perfiles de funcionamiento de pH alterados y/o estabilidad térmica alterada en comparación con las precursoras. También se describen composiciones detergentes y de licuefacción de almidón que comprenden las amilasas mutantes, asícomo los métodos de uso de las amilasas mutantes. Las más particularmente preferidas son las alfa-amilasas mutantes de Bacillus licheniformis modificadas en los residuos MET197 o MET15 o en TRP138 o en residuos equivalentes de otras alfa-amilasas de otras fuentes microbiales (Bacillus, Aspergillus).
