Abstract
(KO) 본 개시는 질량 분석법을 사용하여 이온화 및 검출 전에 면역 글로불린의 사전 정제없이 생물학적 샘플로부터 면역 글로불린을 식별 및/또는 정량화하기 위한 재료 및 방법들에 관한 것이다. 예를 들어, 단일 클론 면역 글로불린으로부터 단일 클론 경쇄는 산과 환원제를 포함하는 수성 완충액으로 단일 클론 면역 글로불린을 포함하는 샘플을 희석한 다음 상기 샘플을 알파-시아노-4-하이드록시신나믹산 매트릭스(alpha-cyano-4-hydroxycinnamic acid, CHCA)와 혼합한 후, 매트릭스 보조 레이저 탈착 이온화 - 비행 시간(matrix assisted laser desorption ionization - time of flight, MALDI-TOF) 질량 분석법을 사용하여 관찰될 수 있다. 또 다른 예에서, 무손상 단일 클론 면역 글로불린은 상기 단일 클론 면역 글로불린을 포함하는 샘플을 물로 희석한 다음 상기 샘플을 CHCA 매트릭스와 혼합한 후, MALDI-TOF 질량 분석법을 사용하여 샘플에서 관찰될 수 있다.
