PROBLEM TO BE SOLVED: To provide hybrid tRNA/pre-microRNA and tRNA/shRNA molecules and methods of making and using.

SOLUTION: A polynucleotide comprises a tRNA operably linked to a pre-microRNA (pre-miRNA) or short hairpin RNA (shRNA). In the polynucleotide, the tRNA is a methionyl tRNA. In the polynucleotide, the pre-miRNA is selected from pre-miRNA-1291, pre-miRNA-34a, pre-miRNA-125b, pre-miRNA-124, pre-miRNA-27b, and pre-miRNA-22.
