Abstract
(EN) The present invention provides novel engineered polypeptides that support both reverse transcription and DNA amplification in manganese-independent reactions. The present invention also provides methods for amplifying template nucleic acids using such polypeptides. This invention addresses deficiencies in the current state of the art in nucleic acid amplification-based detection of template nucleic acids, especially RNA targets, including deficiencies in detection sensitivity, specificity, enzyme stability, inhibitor tolerance and time to result compared with manganese-dependent thermostable reverse transcriptases and two-enzyme solutions.
(FR) La présente invention concerne de nouveaux polypeptides modifiés qui supportent à la fois la transcription inverse et l'amplification d'ADN dans des réactions indépendantes du manganèse. La présente invention concerne également des procédés d'amplification d'acides nucléiques modèles à l'aide de tels polypeptides. La présente invention traite des déficiences dans l'état actuel de la technique se rapportant à la détection d'acides nucléiques modèles basée sur l'amplification d'acides nucléiques, en particulier de cibles d'ARN, notamment des déficiences dans la sensibilité de détection, la spécificité, la stabilité enzymatique, la tolérance aux inhibiteurs et le temps par comparaison avec des transcriptases inverses thermostables dépendantes du manganèse et des solutions à deux enzymes.
