This invention provides, in one embodiment, a recombinant virus or a recombinant virus library wherein each virus comprises a protein involved in viral attachment or infection, a polypeptide which differs by at least a single amino acid from another peptide or polypeptide in the library, and a modified cleavage site that is proximal to the peptide and the protein, wherein the cleavage site is modified such that a compound mediating cleavage has a reduced binding affinity for it as compared to a non-modified cleavage site. The invention further provides a target of interest complex comprising a protease, a target of interest involved in an intermolecular interaction, and a flexible linker that attaches the protease and target of interest. The invention further provides uses thereof, including a method for identifying a peptide which has an intermolecular interaction with a target of interest or identifying the agonistic or antagonistic feature of a peptide that has an intermolecular reaction with a target of interest.

L'invention concerne, dans un mode de réalisation, un virus recombinant ou une bibliothèque de virus recombinants. Chaque virus comprend une protéine impliquée dans une fixation ou infection virale, un polypeptide qui diffère par au moins un acide aminé simple d'un autre peptide ou polypeptide dans la bibliothèque, et un site de clivage modifié à proximité du peptide et de la protéine, ledit site de clivage étant modifié de façon qu'un clivage à composé intercalaire présente une affinité de liaison réduite comparé à un site de clivage non modifié. L'invention concerne en outre une cible d'un complexe d'intérêt comprenant une protéase, une cible d'intérêt impliquée dans une interaction intermoléculaire, et un lieur souple qui se fixe à la protéase et la cible d'intérêt. L'invention concerne également les utilisations associées, y compris un procédé permettant d'identifier un peptide qui a une interaction intermoléculaire avec une cible d'intérêt ou pour identifier la caractéristique agoniste ou antagoniste d'un peptide présentant une réaction intermoléculaire avec une cible d'intérêt.
