Abstract:
(EN) Novel alpha-amylase mutants derived from the DNA sequences of naturally occurring or recombinant alpha-amylases are disclosed. The mutant alpha-amylases, in general, are obtained by $i(in vitro) modifications of a precursor DNA sequence encoding the naturally occurring or recombinant alpha-amylase to generate the substitution (replacement) or deletion of one or more oxidizable amino acid residues in the amino acid sequence of a precursor alpha-amylase. Such mutant alpha-amylases have altered oxidative stability and/or altered pH performance profiles and/or altered thermal stability as compared to the precursor. Also disclosed are detergent and starch liquefaction compositions comprising the mutant amylases, as well as methods of using the mutant amylases. More particularly preferred are mutant alpha-amylases from $i(Bacillus licheniformis) modified at MET197 or MET15 or at TRP138 residues or at equivalent residues of other alpha-amylases from other microbial sources ($i(Bacillus, Aspergillus)).
(FR) Nouvelles alpha-amylases mutantes dérivées de séquences d'ADN d'alpha-amylases naturelles ou de recombinaison. Les alpha-amylsases mutantes, en général, sont obtenues par des modifications in vitro d'une séquence d'ADN précurseur codant l'alpha-amylase naturelle ou de recombinaison pour réaliser la substitution (le remplacement) ou la délétion d'un ou plusieurs restes aminoacides oxydables dans la séquence d'aminoacide d'une alpha-amylase précurseur. De telles alpha-amylases mutantes présentent une stabilité modifiée à l'oxydation et ou des profils modifiés de rendement par rapport au pH et/ou une stabilité thermique modifiée par rapport au précurseur. L'invention porte également sur des compositions de détergents et des compositions de liquéfaction de l'amidon renfermant ces amylases mutantes, ainsi que sur des procédés d'utilisation de telles amylases. Les alpha-amylases mutantes plus particulièrement préférées sont celles provenant de l'espèce $i(Bacillus Licheniformis) modifiée en MET197 ou MET15 ou au niveau des restes TRP138 ou équivalents d'autre alpha-amylases provenant d'autres sources microbiennes $i((Bacillus, Aspergillus)).
