Abstract
(EN) Provided in the present invention are a highly specific glycosyltransferase for rhamnose, and the use thereof. A specific glycosyltransferase is disclosed in the present invention for the first time, which glycosyltransferase can catalyze rhamnosylation at a specific position of a substrate, and has high catalytic activity. Specifically, the specific glycosyltransferase of the present invention can specifically and efficiently catalyze the glycosylation of the C-6 position of a tetracyclic triterpene compound substrate on a first glycosyl so as to extend a rhamnose group. Also provided in the present invention is a mutant of the specific glycosyltransferase. The specific glycosyltransferase of the present invention has good specificity and high efficiency, can be applied to the construction of artificially synthesized ginsenoside, various new ginsenosides and derivatives thereof, and has good application value in the fields of pharmacy, etc.
(FR) La présente invention concerne une glycosyltransférase hautement spécifique pour le rhamnose, et son utilisation. Une glycosyltransférase spécifique est décrite dans la présente invention pour la première fois, ladite glycosyltransférase pouvant catalyser la rhamnosylation à une position spécifique d'un substrat, et possédant une activité catalytique élevée. Plus particulièrement, la glycosyltransférase spécifique de la présente invention peut catalyser spécifiquement et efficacement la glycosylation de la position C-6 d'un substrat de composé triterpénique tétracyclique sur un premier glycosyle de manière à étendre un groupe rhamnose. La présente invention concerne également un mutant de la glycosyltransférase spécifique. La glycosyltransférase spécifique de la présente invention présente une bonne spécificité et une efficacité élevée, peut être appliqué à la construction de ginsénoside synthétisé artificiellement, de divers nouveaux ginsénosides et de leurs dérivés, et a une bonne valeur d'application dans les domaines de la pharmacie, etc.
(ZH) 本发明提供了一种鼠李糖高度专一的糖基转移酶及其应用。本发明首次揭示一种专一性糖基转移酶,其可催化底物特定位置发生鼠李糖基化,具有高的催化活性。具体地,本发明的专一性糖基转移酶能特异和高效地催化四环三萜化合物底物的C-6位在第一个糖基上发生糖基化,以延伸鼠李糖基团。本发明也提供了所述专一性糖基转移酶的突变体。本发明的专一性糖基转移酶具有良好的特异性和高效性,可应用于构建人工合成人参皂苷及多种新人参皂苷及其衍生物,在药学等领域具有良好的应用价值。
