Abstract
(EN) Provided are a new IL-18 substitute and the use thereof. Specifically, provided is a class of nanobodies targeting an IL-18 receptor. The IL-18Rb nanobody and the IL-18Ra nanobody specifically bind to an IL-18Rb chain and IL-18Ra chain of an IL-18 receptor, respectively, and a bispecific antibody based on the two monospecific nanobodies can effectively stimulate PBMC to release IFNγ, and exhibit a significant and effective tumor inhibitory effect. Therefore, the provided antibody can be used as a therapeutic long-acting substitute factor for IL-18, and provides a new strategy for clinical disease treatment.
(FR) L'invention concerne un nouveau substitut d'IL-18 et son utilisation. Plus particulièrement, l'invention concerne une classe de nanocorps ciblant un récepteur d'IL-18. Le nanocorps anti-IL-18 Rb et le nanocorps anti-IL-18Ra se lient de manière spécifique à une chaîne d'IL-18Rb et une chaîne d'IL-18Ra d'un récepteur d'IL-18, respectivement, et un anticorps bispécifique basé sur les deux nanocorps monospécifiques peut stimuler efficacement le PBMC pour libérer l'IFNγ, et présenter un effet inhibiteur tumoral significatif et efficace. Par conséquent, l'anticorps fourni peut être utilisé en tant que facteur de substitution à action prolongée thérapeutique pour IL-18, et fournit une nouvelle stratégie pour le traitement d'une maladie clinique.
(ZH) 提供了新型IL-18替代物及其用途。具体地,提供了一类靶向IL-18受体的纳米抗体。所述IL-18Rb纳米抗体和IL-18Ra纳米抗体分别特异性结合IL-18受体的IL-18Rb链和IL-18Ra链,并且基于两种单特异性纳米抗体的双特异性抗体能够有效刺激PBMC释放IFNγ,且展现出显著有效的肿瘤抑制效果。因此,提供的抗体能够作为IL-18的治疗性长效替代因子,为临床疾病治疗提供新策略。
