Provided are hybrid tRNA/pre-microRNA and tRNA/shRNA molecules and methods of making and using. Accordingly, in one aspect, polynucleotides comprising a tRNA operably linked to a pre-microRNA (pre-miRNA) or short hairpin RNA (shRNA) are provided. In varying embodiments, the tRNA is a methionyl tRNA. In varying embodiments, the pre-miRNA (or shRNA) is selected from premiRNA-1291, pre-miRNA-34a, pre-miRNA- 125b, pre-miRNA- 124, pre-miRNA-27b, and pre-miRNA-22.

L'invention concerne des molécules hybrides d'ARNt/pré-microARN et d'ARNt/ARNsh ainsi que des procédés de fabrication et d'utilisation correspondants. En conséquence, dans un aspect, elle se rapporte à des polynucléotides comprenant un ARNt lié fonctionnellement à un pré-microARN (pré-miARN) ou à un petit ARN en épingle à cheveux (ARNsh). Dans divers modes de réalisation, l'ARNt est le méthionyl-ARNt. Dans divers modes de réalisation, le pré-miARN (ou ARNsh) est choisi parmi le pré-miARN-1291, le pré-miARN-34a, le pré-miARN-125b, le pré-miARN-124, le pré-miARN-27 b et le pré-miARN-22.
