The invention relates to methods for characterizing the binding interactions between binding partners. The invention pertains to methods comprising contacting a first binding partner with a second binding partner that exhibits fast-off rate binding characteristics with the first binding partner to generate the binding interaction between the first binding partner and the second binding partner, wherein the first binding partner is immobilized onto a substrate that is designed to enhance the fluorescence signal of fluorescent molecules located near the substrate's surface, and wherein the second binding partner is a molecule that emits a fluorescent signal or is conjugated to molecule that emits a fluorescent signal. The interactions between the two binding partners can be analyzed based on multiple transient interactions between the two binding partners.

L'invention concerne des procédés de caractérisation des interactions de liaison entre des partenaires de liaison. L'invention se réfère à des procédés consistant à mettre en contact un premier partenaire de liaison avec un second partenaire de liaison, lequel présente des caractéristiques de liaison à vitesse d'arrêt rapide avec le premier partenaire de liaison, afin de générer l'interaction de liaison entre le premier partenaire de liaison et le second partenaire de liaison, le premier partenaire de liaison étant immobilisé sur un substrat qui est conçu pour améliorer le signal de fluorescence de molécules fluorescentes se situant à proximité de la surface du substrat, et le second partenaire de liaison étant une molécule qui émet un signal fluorescent ou est conjugué à une molécule émettant un signal fluorescent. Les interactions entre les deux partenaires de liaison peuvent être analysées sur la base de multiples interactions transitoires entre les deux partenaires de liaison.
