Claims
1. Method for disrupting a biological sample (9), comprising the steps of: ∙ the sample (9) is filled into a non-metallic container (6), ∙ the container (6) is inserted into an adapter (2, 2a), ∙ the adapter, with the sealed container (6) located therein, is connected to an apparatus that automatically reciprocates the adapter, in particular up and down.
2. Method according to claim 1, wherein a plurality of containers (6) with biological samples (9) located therein, which are simultaneously disrupted by the adapter by means the automatic reciprocation, are inserted into the adapter.
3. Method according to any one of the preceding claims, wherein the sample (9) is first washed and then frozen with liquid nitrogen or chemically preserved, and the sample thus prepared is filled into the container (6).
4. Method according to any one of the preceding claims, wherein the adapter comprises casings (4) of metal, into which containers (6) with the biological samples located therein are inserted.
5. Method according to the preceding claim, wherein the containers (6), samples (9) and adapter (2, 2a) are first cooled to less than -20°C, preferably to less than -50°C and the sample
is subsequently disrupted by reciprocating the adapter.
6. Method according to any one of the two preceding claims, wherein the adapter (2, 2a) is cooled not below -80°C.
7. Method according to any one of the preceding claims, wherein at least one moveable hard body (8), preferably two moveable hard bodies, are filled into the container, the bodies consisting of, in particular, metal or glass.
8. Method according to any one of the preceding claims, wherein a lysis buffer is filled into the container (6) prior to disruption, in particular a buffer comprising complexing agents and surfactants for disrupting DNA, preferably ATL or a buffer comprising a chaotropic agent for disrupting RNA, in particular RLT.
9. Method according to any one of the preceding claims, wherein the adapter (2, 2a) is reciprocated at a frequency of at least 10 Hz, preferably of at least 30 Hz.
10. Method according to any one of the preceding claims, wherein the weight of a sample filled into a container (6) is no more than 50 mg, preferably no more than 30 mg.
11. Method according to any one of the preceding claims, wherein the container (6) consists of plastic.
12. Device for carrying out a method according to any one of the preceding claims, comprising an adapter (2, 2a) consisting in particular predominantly of plastic, with casings (4) consisting of metal for accommodating non-metallic containers (6) preferably consisting of plastic.
13. Device according to the preceding device claim, wherein the adapter (2, 2a) is connected detachably, in particular unscrewably, to an apparatus that is capable of automatically reciprocating the adapter parallel to the casings.
14. Device according to the preceding claim, wherein the apparatus is configured such that it is capable of reciprocating the adapter at a frequency of at least 10 Hz, preferably of at least 30 Hz.
15. Device according to any one of the preceding device claims, with containers (6) which are inserted into the casings and in which a biological sample (9) and, additionally, a moveable hard body (8) is located, which consists, in particular, of metal or glass.
16. Device according to any one of the preceding device claims, with an internal casing diameter of the metallic casings (4) of no more than 16 mm, preferably of no more than 12 mm.
17. Device according to any one of the preceding claims, with at least six metallic casings (4), preferably with at least twelve metallic casings (4) for accommodating containers (6).
18. Device according to any one of the preceding claims, wherein the casings (4) are retained by an insert (2a) that is removable from the adapter.
19. Device according to any one of the preceding claims with casings (4) which, in a top view, are disposed in the shape of a ring.
20. Device according to any one of the preceding claims, with a lid (3) that can be firmly connected to the adapter (2, 2a).
